Abstract:AIM: To study the mechanism of SiPc(C35H27O5)8 mediated PDT on retinal pigment epithelial cell(RPE).
METHODS: hRPE cells were divided into blank control group, laser group, photosensitizer SiPc(C35H27O5)8 group, PDT 1J group, PDT 2J group. CCK-8, fluorescent microscopes blue excitation were applied in the experimental groups, and active oxygen level of RPE cell mitochondria and RPE cell apoptosis rate were detected by DCFH-DA.
RESULTS: Different concentration photosensitizer \〖SiPc(C35H27O5)8\〗 mediated PDT on RPE had different effects, in which 5 μg/L of concentration had the most serious damage.
CONCLUSION:New photosensitizer SiPc(C35H27O5)8 mediated PDT on RPE has certain damage effect, its possible mechanism is that the increasing RPE cell mitochondria reactive oxygen species level caused by photosensitizer \〖SiPc(C35H27O5)8\〗 mediated PDT results in cell apoptosis.
